PTC Therapeutics' third-quarter report captures both the accelerating commercial momentum of its newly launched Sephience franchise and the near-term cost of rebuilding its pipeline. After years anchored by the Duchenne muscular dystrophy (DMD) products Translarna and Emflaza, the company's growth story has shifted toward Sephience and the royalty stream from Roche's Evrysdi. The $111 million upfront payment for the ST-920 Fabry disease gene therapy, recorded as an acquired IPR&D expense, drove the quarter into a GAAP loss even as revenue surged. In my view, the results serve as a checkpoint on whether rising sales can outpace the reinvestment needed to sustain long-term growth.
Total revenue for the quarter ended September 30, 2026, came in at $358 million, up approximately 70% from $211 million a year earlier and ahead of the roughly $304 million consensus estimate. Product revenue reached a record $276 million, more than double the $131 million reported in the prior-year period. I also checked this using Tickeron’s AI Screener to see how the stock compares to others in the industry.
Sephience contributed $182 million, up more than 20% sequentially, while the DMD franchise generated $74 million, split between Translarna ($49 million) and Emflaza ($25 million). Royalty revenue from Roche's Evrysdi totaled $82 million.
On a GAAP basis, the company reported a net loss of $0.62 per diluted share, compared with diluted earnings of $0.20 per share in the third quarter of 2025. The swing largely reflected the $111 million acquired IPR&D expense for ST-920, although GAAP operating income still improved to $25.1 million from $3 million a year earlier. Management raised full-year 2026 total revenue guidance to $1.29 billion–$1.32 billion and product revenue guidance to $960 million–$990 million.
Investor reaction to the report reflects a tug-of-war between strong top-line execution and an earnings miss driven largely by a one-time acquisition charge. Revenue beat expectations by a wide margin, and the company raised its full-year guidance, signaling confidence in Sephience's trajectory. At the same time, the GAAP loss of $0.62 per share fell well short of consensus estimates near $0.30, a gap that captures the distorting effect of the ST-920 IPR&D expense rather than underlying operating weakness. Sentiment heading into and around the print has also been shaped by the broader biotech tape, with PTCT shares having declined roughly 17% since the start of the year even as the company's fundamentals improved.
Looking ahead, the central question is whether Sephience can sustain its pace of adoption and carry the company to cash flow break-even. Management described the product as entering a steady, durable growth phase, with the vast majority of the estimated U.S. patient population still untreated. Investors will watch sequential revenue growth, the pace of new patient starts, and discontinuation trends as early signals of the franchise's durability.
International expansion represents a second catalyst. Pricing decisions in Germany and launches in France, Italy, and Spain are expected to shape 2027 growth, making regulatory and reimbursement timelines an important variable.
On the pipeline side, the ST-920 Fabry disease gene therapy is a focal point. A rolling BLA (Biologics License Application) submission is expected to be completed in the fourth quarter of 2026, with potential approval and launch in 2027. Separately, the DMD franchise faces pressure from a new Emflaza generic entrant and the absence of expected large government orders in the fourth quarter, which management has already incorporated into its guidance. Cash of $2.13 billion provides flexibility for additional business development, but rising debt and execution risk around pipeline milestones remain factors worth monitoring.
From what I see, incorporating AI-driven screening has become a regular part of my process for tracking earnings movers like PTCT. Tickeron’s AI Screener helps filter the market using technical patterns, fundamentals, and AI signals, surfacing potential setups across sectors more efficiently than manual methods alone. It’s a practical way to stay organized when monitoring multiple opportunities.
