Amylyx Pharmaceuticals, Inc. (AMLX) is a Cambridge, Massachusetts-based clinical-stage biopharmaceutical company focused on endocrine and neurodegenerative diseases. The stock surged 33.08% to $28.52 in Tuesday morning trading, up from a previous close of $21.43 on Monday, after the company announced that its lead investigational drug, avexitide, met the primary and all secondary endpoints in a pivotal Phase 3 clinical trial.
The sharp rally in AMLX was driven by topline data from the LUCIDITY trial, a 78-participant, multicenter, randomized, double-blind, placebo-controlled study evaluating avexitide in adults with post-bariatric hypoglycemia following Roux-en-Y gastric bypass surgery.
The trial met its FDA-agreed primary endpoint, demonstrating a 55% reduction in the composite rate of Level 2 and Level 3 hypoglycemic events compared with placebo through Week 16, with a p-value of 0.000003. LUCIDITY also met all secondary endpoints, showing consistent and clinically meaningful reductions in Level 2 hypoglycemic events measured by self-monitoring of blood glucose, Level 2 events measured by continuous glucose monitoring, and independently adjudicated Level 3 events.
Safety data supported the positive efficacy readout. Avexitide was generally well-tolerated, with no treatment-related serious adverse events. The most common side effects were diarrhea, injection-site erythema, and injection-site bruising, and no body weight changes were observed in either the treatment or placebo group.
Avexitide is an investigational, first-in-class GLP-1 receptor antagonist designed to block the exaggerated GLP-1-driven insulin response that causes recurrent, dangerous low-blood-sugar episodes in PBH. The condition affects an estimated 160,000 people in the United States, and no FDA-approved treatment currently exists.
The drug holds FDA Breakthrough Therapy and Orphan Drug designations, and Amylyx said it plans to submit a New Drug Application by the end of 2026. If approved, avexitide could become the first treatment for PBH, with a potential commercial launch targeted for 2027. That regulatory visibility added to the positive market reaction. Earlier this week, Mizuho raised its price target on the stock to $30 from $24 while maintaining an Outperform rating, citing higher launch-price assumptions for avexitide.
The move in AMLX stood out against a cautious broader tape. Major equity indexes opened under modest pressure Tuesday, underscoring that the advance was driven by company-specific clinical news rather than sector or macroeconomic tailwinds. The biotech complex was also mixed, making the outperformance especially notable.
Trading volume was elevated in early action as investors repriced the binary clinical outcome. The stock broke decisively above its recent trading range and prior 52-week highs, reinforcing the technical significance of the session's move.
Attention now shifts to execution. The company is preparing an NDA submission for avexitide by year-end, with the LUCIDITY open-label extension and an expanded access program still ongoing. Full trial data are expected to be presented at an upcoming medical meeting, which could provide additional detail on secondary endpoints and longer-term safety.
Risks remain, including FDA review uncertainty, the demands of a potential 2027 commercial launch, and the inherent volatility of clinical-stage biotech stocks after large binary moves. Amylyx has previously stated that its cash position is expected to fund operations into 2028, giving it runway to advance regulatory and prelaunch activities.
